    Exclamation Kefir ovvero l'Elisir di Lunga Vita

    Diversi studi hanno evidenziato le proprietà miracolose del Kefir, ovvero del latte fermentato per circa 24 ore a temperatura ambiente mediante i grani di Kefir.

    Diversi articoli e sperimentazioni pubblicate su PubMed evidenziano che i batteri e i lieviti contenuti nei grani di Kefir sono in grado in particolare di attivare il sistema immunitario e le cellule killer che in tal modo riescono a neutralizzare più facilmente microrganismi patogeni e cellule tumorali.

    Si ritiene che l'origine di tale composto si probiotici e lieviti che vivono in simbiosi si siano originati grazie dalle usanze dei pastori del Caucaso che erano soliti riporre il latte nello stomaco essiccato delle capre.

    E guarda caso tra tutti gli animali esistenti le capre sono l'unica specie animale nella quale i tumori sono rarissimi.

    Antitumor Effects
    Kefir also has significant antitumor activity against multiple cancer cell types. L. kefiri was shown to increase apoptosis of multiple drug resistant human myeloid leukemia cells in vitro through the activation of caspase 3 in a dose dependent manner (Ghoneum and Gimzewski, 2014). The cell free fraction of kefir has shown antitumor activity in vitro when it was observed to have a dose dependent anti-proliferative effect on the gastric cancer cell line SGC7901 (Gao et al., 2013). This study further demonstrated that cell free kefir was able to induce apoptosis in SGC7901 cells through up regulation of the bax gene, and apoptosis promoter and anti-oncogene, and down regulation of the bcl-2 gene, which is an apoptosis inhibitor and known oncogene (Sorenson, 2004). In addition to the promotion of cell death in cancerous cells, antimutagenic effects have been demonstrated in studies with known carcinogens such as methylmethanosulphate, methy-lazoxymethanol, sodium azide, aflatoxin B1, and 2-aminoanthracene as indicated by the Ames test (Guzel-Seydim et al., 2006).

    In mouse models of fusiform cell sarcomas, mice receiving intraperitoneal kefir had reduced tumor size, with some tumors completely disappearing over a 20 days treatment period (Cevikbas et al., 1994). While this is impressive, it has yet to be determined if these findings can be replicated in the case of oral consumption. A separate study utilizing a murine breast cancer model showed that kefir feeding prior to challenge with the tumor resulted in decreased size and increased apoptosis of the tumor, and that the levels of IgA+ cells and CD4+ T cells were also increased (de Moreno de LeBlanc et al., 2007). Mice with breast cancer tumors fed kefir also showed increased serum levels of Il-10 and IL-4 (de Moreno de LeBlanc et al., 2006). These studies both showed increases in immune cell populations and recruitment, pointing to a possible mechanism for the reduction of tumor size. These findings are consistent with other studies that have shown that kefir is able to modulate the immune system in the gut and show that the immunomodulatory abilities of kefir may not be limited to the gastrointestinal tract (Thoreux and Schmucker, 2001; Vinderola et al., 2005; Correa Franco et al., 2013).

    Abstract
    The effects of oral administration of milk and soy milk kefirs on tumor growth in tumor-bearing mice and the mucosal immunoglobulin A response in mice were studied. Oral administration of milk and soy milk kefirs to mice inoculated with sarcoma 180 tumor cells resulted in 64.8% and 70.9% inhibition of tumor growth, respectively, compared with controls. In addition, oral administration of the two kefir types induced apoptotic tumor cell lysis. Total immunoglobulin A levels for tissue extracts from the wall of the small intestine were also significantly higher for mice fed a milk kefir or a soy milk kefir regimen for 30 days. These results suggest that milk and soy milk kefirs may be considered among the more promising food components in terms of cancer prevention and enhancement of mucosal resistance to gastrointestinal infection.

    Elucidating the Immune-Related Mechanisms by Which Probiotic Strain Lactobacillus casei BL23 Displays Anti-tumoral Properties.

    Abstract
    We have recently described antitumor properties of Lactobacillus casei BL23 strain in both a mouse allograft model of human papilloma virus (HPV)-induced cancer and dimethylhydrazine-associated colorectal cancer. However, the mechanisms underlying these beneficial effects are still unknown. Interestingly, in vitro cellular models show that this bacterium is able to stimulate the production of high levels of IL-2. Because this cytokine has well-known antitumor properties, we decided to explore its role in the anti-cancer effects of BL23 using the HPV-induced cancer model. We found a negative correlation between IL-2 and tumor size confirming the necessity of IL-2 to protect from tumor development. Then, we blocked IL-2 synthesis using neutralizing monoclonal antibodies in mice that were challenged with lethal levels of tumor cells; this led to a significant reduction in the protective abilities of BL23. Next, we used a genetically modified strain of Lactococcus lactis to deliver exogenous IL-2 to the system, and in doing so, we were able to partially mimic the antitumor properties of BL23. Additionally, we showed the systemic role of T-cells in tumor protection through a negative correlation between tumor size and T-cells subpopulations and an increasement of BL23-specific local Foxp3 levels in tumor-bearing mice. Finally, we observed a negative correlation between tumor size and NK+ cells, but local recruitment of NK cells and cytotoxic activity appeared specific to BL23 treatment. Taken together, our data suggest that IL-2 signaling pathway plays an important role in the anti-tumoral effects of probiotic strain L. casei BL23. These results encourage further investigation in the use of probiotic strains for potential therapeutic applications to clinical practice, in particular for the treatment of colorectal cancer. Furthermore, our approach could be extended and applied to other potential beneficial microorganisms, such as gut microbiota, in order to better understand the crosstalk between microbes and the host.
